So, I've noticed how the nurse sometimes jokes around about being gay, and how Emi takes it so seriously and offensively like it has happened before. If you remember Emi said that the track captain is Gay and had a crush on him until she found out. You guy's see where I'm going with this?

The Nurse does assure Hisao that he's ABSOLUTELY SURE that Emi and the team captain are most DEFINITELY not in a relationship...

I think this is a stretch.
The nurse has his fun, but he is wholly professional when matters are serious and you can't enter relationships with patients as it's a conflict of interest.

I think you're reading into it a bit much. I think she burst in and called the nurse an asshole because she knew his joke meant that they were done. "Asshole" almost seems to be Emi's term of endearment for the nurse.

i have the theory who Emi is a bit fujoshi-like too (she is already a bit perverted and sensual, this is only something to add) and Nurse, who knows her for years, does all that jokes for make fun with her.
And about the track Capitain, she is more jolly about the stuff who he tell her about the "fun" in the in the back storage (oh another pun) than the deseption about her discover about his sexuality, who makes me think more about her fujoshi behaviour.
